毒性Intercalating dye widely used to stain DNA in gels and gradients. The DNA can be visualized readily by irradiation with ultraviolet light, as little as 0.5 μg of DNA being detectable by such methods. Ethidium bromide is also added to cesium chloride density gradients, since as an intercalating molecule it binds more readily to linear DNA than to closed collinear circles of DNA (such as plasmids). Binding of ethidium bromide reduces the density of DNA, thus covalent circles of DNA have higher densities at saturating concentrations of ethidium bromide, permitting the separation of plasmid DNA. Ethidium bromide is a putative carcinogen.

溴化乙錠是一種高度靈敏的熒光染色劑,用于觀察瓊脂糖和聚丙烯酰胺凝膠中的DNA。溴化乙錠用標(biāo)準(zhǔn)302nm 紫外光透射儀激發(fā)并放射出橙紅色信號,可用Polaroid 底片或帶CCD 成像頭的凝膠成像處理系統(tǒng)拍攝。

熒光探針技術(shù)中的熒光探劑
溴化乙啶(簡稱EB)是目前在紫外線下觀察DNA而常用的一種熒光染料,屬于插入染料,原因是它有多環(huán)結(jié)構(gòu),這種結(jié)構(gòu)使它能夠插入核苷酸堿基之間。由于核酸和堿基的熒光量子產(chǎn)率很低,因此利用天然熒光檢測核酸比蛋白質(zhì)困難更大,所以熒光探針技術(shù)在核酸研究中顯得尤為重要。比較常用的是溴化乙啶(et-hidium bromide或稱菲啶溴紅),它與核酸 (DNA、RNA、雙鏈多聚核苷酸)有特異結(jié)合的能力。它對脫氧核糖核酸的最低檢測量可達(dá)10ng/ml,而且它與核酸的雙鏈區(qū)結(jié)合是專一的,利用它與各種構(gòu)象核酸結(jié)合比例的不同所產(chǎn)生的熒光強(qiáng)度的變化,可以把各種構(gòu)型核酸加以區(qū)分。例如它能鑒別天然核酸和變性核酸,區(qū)分線性DNA、環(huán)狀DNA和超級線團(tuán)DNA等。此外各種吖啶,包括吖啶橙、吖啶黃以及前黃素等都是核酸常用的探劑。
熒光分析法可分為直接測定法和間接測定法二種。直接測定法是利用物質(zhì)自身發(fā)射的熒光,即所謂“自熒光”或“內(nèi)源熒光”來進(jìn)行測定。但在自然界中只有少部分物質(zhì)在光激發(fā)后可以發(fā)射熒光,而有相當(dāng)一部分物質(zhì)發(fā)熒光很弱或不發(fā)射熒光,如蛋白和核酸就是發(fā)熒光很弱的物質(zhì)。為了充分利用熒光分析法靈敏度高這一特點(diǎn)就需使其轉(zhuǎn)化成熒光物質(zhì)再進(jìn)行測定。此時(shí)可用某些試劑(如熒光染料) 使其與熒光較弱或不顯熒光的物質(zhì)共價(jià)或非共價(jià)結(jié)合,以形成發(fā)熒光的結(jié)合物再進(jìn)行測定,這就是所謂的“熒光探針”技術(shù)或“熒光標(biāo)記”技術(shù)。使試劑(熒光劑) 吸附在大分子上或共價(jià)結(jié)合到大分子上。所用的染料溴化乙啶被叫做“熒光探劑”。探劑熒光特性的變化,實(shí)際上是由于大分子構(gòu)象的變化所引起的探劑環(huán)境變化的信號,所以利用這種信號還可以得到大分子構(gòu)象變化的信息。

Ethidium Bromide (Homidium bromide, EtBr, EB) 是一種嵌入劑,與DNA堿基對類似。EtBr在分子生物學(xué)實(shí)驗(yàn)技術(shù)(如瓊脂糖凝膠電泳)中經(jīng)常被用于熒光標(biāo)記(核酸染色)。
體外研究

Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
The preparation of agarose gel:
1. Agarose gels are commonly used in concentrations of 0.5% to 2.5% depending on the size of bands needed to be separated.
2. Mix the agarose powder with 1X TAE/TBE.
3. Microwave for 1-3 min until the agarose is completely dissolved ( Caution : not overboil).
4. Make the solution cool down before solidification.
5. Add ethidium bromide (EtBr) to a final concentration of approximately 0.2-0.5 μg/mL (Stocks are generally 10 mg/ml, and require 5 μL stock/100 mL gel).
6. Ethidium bromide binds to the DNA and you could visualize the DNA under ultraviolet (UV) light.
CAUTION: EtBr is a known mutagen. Please pay attention to strengthening protection.
