Stage #2: with potassium carbonate in water; for 16 h;Reflux;

1.b b. PREPARATION OF 4-ETHYL-5-(PYRIDIN-4-YL)-4H-1,2,4-TRIAZOLE-3-THIOL

[00446] To a solution of isonicotinohydrazide (84 mg, 0.61 mmol) in 1.0 mL of ethanol was added ethyl isothiocyanate (64 μ, 0.74 mmol). This reaction mixture was heated in a microwave reactor for 15 min at 150°C, cooled to room temperature and concentrated. The residue was then re-dissolved 10 ml of H₂Oand K2CO₃ (101.5 mg, 0.74 mmol) was added, then the solution was brought to reflux. After 16 h,the reaction was allowed to cool to room temperature, diluted with methanol and concentrated. The residue was purified by column chromatography with methanol/CH₂Cl2 (1 :6) to afford 67 mg (53%) of the title compound. ^lH NMR (MeOD) 58.78 (d, J = 5.5 Hz, 2H), 7.78 (d, J = 6.2 Hz, 2H), 4.26 (q, J = 7.2, 2H), 1.33 (t, J = 7.3 Hz, 3H)LRMS calculated for C₉Hi₀N₄S (M+H)Vz: 207.06. Measured 207.1 m/z.
