名稱	Gramicidin
描述	Gramicidin (Gramicidine) is an antimicrobial peptide. Gramicidin dimers form ion channel-like pores in the cell membranes and organelles of bacterial and animal cells, allowing inorganic monovalent ions, such as potassium and sodium ions, to pass freely through the pores by diffusion, disrupting the important ionic gradient between membranes and thus killing the cells by various actions. Gramicidin is effective against Gram-positive bacteria such as Bacillus subtilis and Staphylococcus aureus, but is not as effective against Gram-negative bacteria such as Escherichia coli.
體外活性	方法: OC 細(xì)胞 OVCAR8、A2780 和 SKOV3 用 Gramicidin (0.33-3 μM) 處理 24-72 h，通過 WST-1 assay 檢測細(xì)胞活力。結(jié)果: Gramicidin 以劑量和時間依賴的方式抑制 OC 細(xì)胞的生長，72 h 時 OVCAR8、SKOV3 和 A2780 細(xì)胞的 IC50 值分別為 0.0763、0.1856 和 0.1148 μM。[1] 方法: 人胃癌細(xì)胞 SGC-7901 用 Gramicidin (0.3-3 μM) 處理 24-48 h，通過 Flow cytometry 檢測細(xì)胞凋亡。結(jié)果: Gramicidin 以時間和劑量依賴的方式促進(jìn) SGC-7901 細(xì)胞的凋亡。當(dāng)細(xì)胞與 Gramicidin 一起培養(yǎng) 48 h 時，與對照組相比，3 μM Gramicidin 處理的活細(xì)胞百分比降至 54.2%。[2]
體內(nèi)活性	方法: 為研究對體內(nèi)同種異體移植物存活的影響，將 Gramicidin (2-4 mg/kg) 腹腔注射給異位心臟移植大鼠，每天一次，持續(xù) 6 天。結(jié)果: 對照組的移植物搏動在移植后 5.4+/-0.4 天因急性同種異體排斥反應(yīng)而停止，而接受 Gramicidin (2-4 mg/kg) 腹腔注射組的受者移植物搏動分別顯著延長至移植后 15.4+/-4.3 或 18.4+/-3.9 天。[3]
