Rilonacept，也被稱為 IL-1 Trap，是一種重組融合蛋白，由人 IL-1 受體 I 型（IL-1RI）的胞外結構域和人 IgG1 的 Fc 段組成。它在哺乳動物細胞表達系統(tǒng)中產(chǎn)生，能廣泛地在多種免疫相關細胞環(huán)境中發(fā)揮作用，在單核細胞、巨噬細胞等免疫細胞表面有較高的親和力結合位點。當機體遭遇炎癥刺激時，Rilonacept 可迅速與炎癥關鍵介質(zhì)白細胞介素 - 1α（IL-1α）和白細胞介素 - 1β（IL-1β）緊密結合，阻斷其與細胞表面天然受體的相互作用，從而有效抑制 IL-1 介導的炎癥信號傳導通路。在臨床試驗和基礎研究中發(fā)現(xiàn)，Rilonacept 可以顯著降低多種自身炎癥性疾病動物模型的炎癥指標，如在家族性寒冷自身炎癥綜合征模型中，它能明顯減輕發(fā)熱、皮疹和關節(jié)疼痛等癥狀；在痛風性關節(jié)炎的研究中，它也表現(xiàn)出對炎癥發(fā)作的有效抑制，減少關節(jié)腫脹和疼痛程度，這表明它在炎癥相關疾病的發(fā)病機制研究以及新型抗炎藥物研發(fā)方面具有極高的研究價值。
