背景（Background）

基質(zhì)金屬蛋白酶-2（MMP-2）也稱為72 kDa IV型膠原酶、72 kDa明膠酶、明膠酶A和CLG4A，屬于肽酶M10A家族。MMP-2/CLG4A含有3個(gè)纖維連接蛋白II型結(jié)構(gòu)域和4個(gè)血紅素樣結(jié)構(gòu)域。MMP-2由正常皮膚成纖維細(xì)胞產(chǎn)生。MMP-2切割膠原樣序列Pro-Gln-Gly-|-Ile-Ala-Gly-Gln。MMP2參與多種功能，如血管重塑、血管生成、組織修復(fù)、腫瘤侵襲、炎癥和動(dòng)脈粥樣硬化斑塊破裂。除了降解細(xì)胞外基質(zhì)蛋白外，還可以作用于幾種非基質(zhì)蛋白，如大內(nèi)皮1和β型CGRP，促進(jìn)血管收縮。還可以通過Gly-|-Leu鍵切割KISS。似乎在心肌細(xì)胞死亡途徑中起作用。通過調(diào)節(jié)GSK3β的活性導(dǎo)致心肌氧化應(yīng)激。體外切割GSK3β。PEX是MMP2的C末端非催化片段，具有抗血管生成和抗腫瘤特性，并抑制細(xì)胞遷移和細(xì)胞與FGF2和玻連蛋白的粘附。
