描述	D-Luciferin potassium (D-Luciferin K Salt) is a luciferase (Luc) substrate that catalyzes the production of typical yellow-green light (560 nm) by bioluminescent insects. D-Luciferin potassium can be used in luciferase assays.
激酶實驗	Enolase activity assay is in the 2.0 mM MgSO4 and 400 mM KCl in the absence or presence of ENOblock or NaF, at 37°C by incubating pure enolase (3–9 U) in a buffer containing 50 mM imidazole-HCl (pH 6.8). The reaction is initiated by adding 1 μmol of 2-phospho-D-glycerate, and the OD is measured after 10 min of reaction time with a spectrophometer at 240 nm.
體外活性	方法：用于體外生物發(fā)光分析的 D-Luciferin potassium 的制備： 1、用無菌水配制 200× D-Luciferin 儲備液 (30 mg/mL)，輕輕混勻，直到全部溶解。配制完成后可立即使用，也可分裝儲存于 -20℃。 2、在預(yù)熱的組織培養(yǎng)基中配制 1× D-Luciferin (150 μg/mL) 工作液。 3、吸出培養(yǎng)細(xì)胞的培養(yǎng)基。 4、在成像前向細(xì)胞中加入 1× D-Luciferin 溶液。成像前將細(xì)胞在 37℃ 下培養(yǎng)一小段時間可增加信號。
體內(nèi)活性	方法：用于體內(nèi)生物發(fā)光分析的 D-Luciferin potassium 的制備： 1、用 DPBS 配制新鮮的 15 mg/mL D-Luciferin 儲備液。用 0.20 um 過濾器過濾消毒。 2、按小鼠體重注射 10 μL/g。每只小鼠應(yīng)注射 150 mg D-Luciferin/kg 體重。例如，對于體重 10 g 的小鼠，注射 100 μL 以提供 1.5 mg D-Luciferin。 3、在成像前 10-15 min 腹腔注射 D-Luciferin。應(yīng)為每種動物模型進(jìn)行熒光素動力學(xué)研究，以確定峰值信號時間。
