Integrin αvβ8分子背景
整合素αVβ8（ITGAV和ITGB8或ITGAVB8）在卵黃囊、胎盤、腦血管周圍星形膠質(zhì)細胞、雪旺氏細胞、腎小球系膜細胞和肺上皮細胞中表達。與其他αV整合素不同，ITGAVB8似乎沒有呈現(xiàn)不同的激活狀態(tài)，細胞質(zhì)尾部也沒有連接到細胞骨架。它結(jié)合含有RGD基序的配體，包括玻連蛋白、纖維蛋白和潛在TGF-β復合物的潛伏相關(guān)肽（LAP）。αVβ8與LAP的高親和力結(jié)合允許MT1-MMP進行蛋白水解切割，從而釋放活性TGF-β。這種機制不同于αVβ6，另一種αV整合素可以通過非蛋白水解機制從潛伏期激活TGF-β。TGF-β激活的下游效應包括控制細胞生長和相關(guān)的血管化。
