IL-2分子背景
白細(xì)胞介素-2（IL-2）是一種白介素，一種細(xì)胞因子免疫系統(tǒng)信號(hào)分子，是一種促白細(xì)胞激素，在身體對(duì)微生物感染的自然反應(yīng)中起著重要作用，并有助于區(qū)分外來（非自身）和自身。IL-2通過與淋巴細(xì)胞（負(fù)責(zé)免疫的細(xì)胞）表達(dá)的IL-2受體結(jié)合來介導(dǎo)其作用。成熟的人IL-2與小鼠和大鼠IL-2的aa序列同源性分別為56%和66%。人和小鼠IL-2具有跨物種活性。IL-2受體由三個(gè)亞基組成，它們以不同的預(yù)形成復(fù)合物存在于細(xì)胞表面。IL-2在胸腺T細(xì)胞發(fā)育過程中也是必要的，用于成熟被稱為調(diào)節(jié)性T細(xì)胞（T-reg）的獨(dú)特T細(xì)胞亞群。從胸腺中排出后，T-Regs的功能是防止其他T細(xì)胞識(shí)別和對(duì)抗“自身抗原”，這可能會(huì)導(dǎo)致“自身免疫”。T-Regs通過阻止反應(yīng)細(xì)胞產(chǎn)生IL-2來實(shí)現(xiàn)這一目的。因此，需要IL-2來區(qū)分自我和非自我，這是免疫系統(tǒng)的另一個(gè)獨(dú)特特征。
