菜油甾醇是一種植物甾醇，是甾體藥物（孕酮、雄烯二酮、氫化可的松等）的一種重要合成前體，也是植物來源的主要甾醇之一。其與動物來源的主要甾醇（膽固醇）的結構僅在C24位多出一個甲基，與微生物來源的主要甾醇（麥角甾醇）的區(qū)別在于C7~8和C22~23位是飽和單鍵。菜油甾醇可作為一種植物甾醇用于檢測其對乳腺癌細胞中肝臟X受體α(LXRA)轉錄激活的影響。它可在液相色譜多反應監(jiān)測(LC-MRM)分析中作為甾醇標準品用于生成校準曲線。Campesterol是一種植物甾醇，具有降膽固醇和抗癌作用。

Campesterol shows a weak cytotoxicity in non-proliferating human umbilical vein endothelial cells (HUVECs). Within the non-cytotoxic concentration range, campesterol significantly inhibits the bFGF-induced proliferation and tube formation of HUVECs in a concentration-dependent manner, while it does not affect the motility of HUVECs. 50 μg/mL of campesterol decreases the cell viability up to about 56% of control(IC 50 of over 50 μg/mL).
Campesterol effectively disrupts the bFGF-induced neovascularization in chick chorioallantoic membrane (CAM) in vivo .

生產(chǎn)方法
重組釀酒酵母生物合成菜油甾醇：首先通過生物信息學分析，篩選了10種不同來源的7-脫氫膽固醇還原酶DHCR7，并采用CRISPR/Cas9基因編輯技術將釀酒酵母（Saccharomyces cerevisiae）內(nèi)源的ERG5基因替換成不同來源的DHCR7基因，構建了菜油甾醇合成菌株。結果發(fā)現(xiàn)整合來源于Pangasianodon hypophthalmus DHCR7的菌株Zw507表現(xiàn)出最高的菜油甾醇的產(chǎn)量216.93 mg/L。進一步篩選了10種酵母內(nèi)源啟動強度較強的啟動子來與PhDHCR7基因進行組合，結果顯示以TEF1p為啟動子時菜油甾醇的產(chǎn)量最高可達253.35 mg/L。為了進一步提高菜油甾醇產(chǎn)量，增加了DHCR7表達盒在酵母基因組上的拷貝數(shù)。當拷貝數(shù)為3個時，菜油甾醇的產(chǎn)量達到最高302.27 mg/L。最終，通過5 L發(fā)酵罐進行補料分批發(fā)酵，實現(xiàn)了916.88 mg/L菜油甾醇產(chǎn)量。該菌株可作為后續(xù)甾體藥物生物合成的優(yōu)良底盤細胞。釀酒酵母體內(nèi)菜油甾醇生物合成途徑
