Allgemeine Beschreibung
MANF (mesencephalic astrocyte-derived neurotrophic factor) forms a family of neurotrophic factors (NTFs) along with cerebral dopamine neurotrophic factor (CDNF). It is characterized by eight conserved cysteine residues, a saposin-like domain in the N-terminal and a putative unfolded carboxy-terminal domain. This protein has a 21 amino acids long signal peptide, which is cleaved off to yield a mature protein of 158 amino acids. MANF is a secreted protein, but some of it remains in endoplasmic reticulum (ER). It was initially isolated from cultured embryonic dopaminergic neurons, and in brain, is predominantly expressed in cerebral cortex, hippocampus and cerebellar Purkinje cells. This gene is localized to human chromosome 3, spans 4.3kb, and contains 4 exons. The encoded protein has a molecular weight of 20kDa.
Biochem/physiol Actions
MANF (mesencephalic astrocyte-derived neurotrophic factor) plays a protective role towards midbrain dopaminergic neurons. It is an endoplasmic reticulum (ER) stress related protein. Its levels are increased post brain injury, ischemia and epileptic insults. It might confer neuro-protection against brain ischemia, by preventing necrosis or apoptosis of cerebral cortex cells. It is over-expressed during unfolded protein response (UPR), and prevents ER stress-induced apoptosis, as well as cell proliferation.
