Allgemeine Beschreibung
The gene IRAK3 (interleukin 1 receptor associated kinase 3), also referred to as IRAKM, encodes a member of the IRAK family. The members of this family are involved in several inflammatory processes. The encoded protein is selectively expressed in monocytes and macrophages. The gene is mapped to human chromosome 12q14.3.
Biochem/physiol Actions
The members of the IRAK (interleukin 1 receptor associated kinase) family, IRAK-1 and IRAK-4 form a complex with MyD88 (myeloid differentiation primary response 88) and TRAF-6 (TNF receptor associated factor 6), upon sensing of lipopolysaccharide by TLR-4 (Toll like receptor). This complex functions in internal signaling and activates MAPK (mitogen activated protein kinase) and NF (nuclear factor)-κB pathways. The protein encoded by IRAK-M, however, has a negative effect on the complex. It is an inhibitor of IRAK-1/IRAK-4 and negatively regulates Toll-like receptor signaling by inhibiting the dissociation of the complex formed by TLR-4 receptor activation. Mutations in this gene have been associated with the pathogenesis of early-onset persistent asthma.
