Allgemeine Beschreibung
Mediator of DNA damage checkpoint protein 1 (MDC1) is a subunit of the cohesin complex and is an essential protein required for sister chromatid cohesion in mitosis and meiosis. Temperature-sensitive mutations of this protein have been associated with defects in sister chromatid cohesion, chromosome condensation, and chromosome segregation. Its expression is cell cycle regulated and peaks in S phase. Expression of MDC1 is required for the S- and G2/M-checkpoint-mediated cell cycle arrest in response to DNA damage. MDC1 has been shown to interact with H2A.X near sites of DNA double-strand breaks and recruit ATM kinase to the damaged region of DNA. Consistent with this role, MDC1 also appears to modulate the function of other proteins involved in the DNA damage checkpoint such as BRCA1 and Chk2.
