Human G9a (euchromatic histone-lysine
N-methyltransferase 2) with N-terminal GST tag, MW = 74.6 kDa, expressed in an
E. coli expression system. G9a is a member of SET (Su(var)3-9, enhancer-of-zeste and trithorax) domain containing, Su(var)3-9 family of proteins. This domain is evolutionary conserved.
Biochem/physiol Actions
The encoded protein G9a (euchromatic histone-lysine N-methyltransferase 2) is a methyltransferase that methylates lysine residues of histone H3. Overexpression of G9a is observed in cancer. It is known to participate in cancer cell migration, invasion and metastasis. G9a is an epigenetic regulator and can inhibit the action of tumor suppressor genes, thereby, contributes to carcinogenesis. Thus, G9a is considered to be an oncogenic and metastatic factor. It also affects DNA damage repair mechanism, which might make tumor cells resistant to radiation and chemotherapy. G9a serves as a target for cancer therapy including breast cancer.
