Reaction Conditions:
Stage #1: 4-hydroxy-3,5-dichlorobenzoic acidwith potassium carbonate in N,N-dimethyl-formamide at 20; for 0.5 h;
Stage #2: methyl iodine in N,N-dimethyl-formamide at 80; for 3 h;
Steps:
5.1.1. General procedure for the synthesis of compounds 2 and 5
General procedure: 3,5-dichloro-4-hydroxybenzoic acid (5.00g, 24.15mmol) and K2CO3 (6.68g, 48.3mmol) was added into a 200mL flash. The mixture was introduced with DMF (30mL), and was stirred under room temperature for 30min. Then halohydrocarbon (48.31mmol) was added, and the mixture was stirred at 80°C for 3h. After the starting material was completely consumed, water (100mL) was added and the mixture was extracted with ethyl acetate (200mL x 3). The combined extracts were washed with brine, and the solution was removed using rotary evaporation on a water bath. The residue was purified on silica gel column chromatography to yield the desired intermediates.
