Step 4: l,3,9-Trimethyl-7,9-dihydro-lH-purine-2,6,8(3H)-trione: To a magnetically stirred solution of Step 3 intermediate (2.0 g, 10.857 mmol) in anhydrous THF (50.0 mL) was added Ι,Γ-carbonyldiimidazole (2.1 g, 13.029 mmol) at room temperature under nitrogen atmosphere. After refluxing for 18 h the excess of solvent was distilled under reduced pressure. The residue obtained was diluted with ethyl acetate (100 ml) and water (100 ml). The aqueous layer was extracted with ethyl acetate (2 x 75 ml) and the combined organic layers were washed with water (2 x 25 ml) followed by brine (25 ml), dried (Na₂S0₄) and filtered. The filtrate was concentrated under reduced pressure. The residue obtained after the distillation of the solvent was purified by silica gel column chromatography using methanol in chloroform to obtain 1.2 g of the product as a white solid; 1H NMR (300 MHz, DMSO-cfe) δ 3.19 (s, 3H), 3.48 (s, 3H), 3.63 (s, 1H), 1 1.12 (br s, 1H); APCI-MS (m/z) 21 1.21 (M+H)⁺.
