154.5g of 4-chloro-1H-pyrazolo [3,4-d] pyrimidine (1mol) was dissolved in 200mL of ethanol, first added 122g ofTriethylamine (1.2mol), and then slowly dropwise added 138g, a concentration of 28wt% aqueous ammonia (1.1mol), dropping end heated to reflux, the reaction 5h. After the reaction was completed, the ethanol was evaporated to dryness, water was added and the mixture was extracted three times with ethyl acetate. The organic layer was spin-dried to give 130 g of adenine as a white solid in a yield of 96.3%.
