with N-iodo-succinimide in N,N-dimethyl-formamide at 20;Inert atmosphere;

Steps:

36.1 Step 1 :

Under inert atmosphere, to a solution of 6-chloro-4-methyl-pyridin-3-amine (200 mg, 1 .40 mmol) in dry DMF (7 ml.) was added /V-iodo-succinimide (346 mg, 1.54 mmol). The reaction mixture was strirred overnight at room temperature, then water (70 mL) was added. The precipitate was filtered, washed with water and dried overnight under high vacuum in presence of P205 at 50 °C to afford 6-chloro-2-iodo-4- methyl-pyridin-3-amine (265 mg) as a brown solid. The filtrate was then extracted with dichloromethane (2 x 30 mL), dried over MgS04 and concentrated under vacuum. Water was added to the resulting crude residue to precipitate the product which was filtered and dried overnight under high vacuum in presence of P205 at 50 °C to afford more 6-chloro-2-iodo-4-methyl-pyridin-3-amine (160 mg) as an orange solid. In total 425 mg of 6-chloro-2-iodo-4-methyl-pyridin-3-amine were obtained (quantitative yield).
