Steps:
R.39.1 Ethyl 3-methyl-5-nitro-1H-indole-2-carboxylate
To a solution of 3-methyl-5-nitro-1H-indole-2-carboxylic acid (400 mg, 1.82 mmol) in DMF (5 mL), potassium carbonate (755 mg, 5.46 mmol) and ethyl iodide (284 mg, 2.20 mmol) were added, and the mixture was stirred at room temperature for 6 hours.
Water was added to the reaction solution, followed by extraction with ethyl acetate twice.
The organic layer was washed with a saturated saline solution, dried over anhydrous sodium sulfate and filtered, and then the organic layer was concentrated under reduced pressure to obtain the title compound (440 mg, 100%).
