Stage #2: methyl iodide in tetrahydrofuran;diethyl ether at -78 - 20; for 21 h;

1A EXAMPLE 1; 7-((3R)-3-aminopyrrolidin-1-yl)-6-fluoro-5-methyl-4-oxo-1,4-dihydro-1,8-naphthyridine-3-carboxylic Acid; Example 1A; 2,6-dichloro-5-fluoro-4-methylnicotinic Acid and 2,6-dichloro-5-fluoro-4-ethylnicotinic Acid
A solution of 2,6-dichloro-5-fluoronicotinic acid (1.99 g) in THF (40 mL) at -78° C. was treated with 1.4M methyllithium in diethyl ether (14 mL), stirred at -25° C. for 1.5 hours then at -78° C. for one hour, treated with iodomethane (1.4 mL), stirred at 0° C. for 2 hours and -20° C. for 18 hours, treated with water, concentrated to remove the THF, diluted with water, washed with diethyl ether, acidified with 1M HCl, and extracted with diethyl ether; and the extract was dried (MgSO4), filtered, and concentrated.
