Reaction Conditions:

Stage #1:1-phenylmethyl-4-piperidone;methylmagnesium bromide in tetrahydrofuran;diethyl ether at -15 - 20;
Stage #2: with water;ammonium chloride in tetrahydrofuran;diethyl ether

Steps:

Description 1; . 1-Benzyl-4-methylpiperidin-4-ol (D1); ) A stirred solution of 3M methylmagnesium bromide in ether (132ml, 396mmol) was cooled by dry ice-acetone bath to -150C and a solution of 1-benzylpiperidin-4-one (3Og, 158mmol) in anhydrous tetrahydrofuran (300ml) was added dropwise at -150C. The mixture was stirred for 2h, the temperature rose to room temperature and the mixture was stirred overnight. After that, the mixture was quenched with saturated ammonium chloride solution (200ml) and the mixture was evaporated to remove tetrahydrofuran. The residue was diluted with ethyl acetate (2 x 500 ml), washed with saturated aqueous sodium bicarbonate solution (500ml), water and brine, then dried over sodium sulfate and concentrated in vacuo to give the title compound (26g, 80%).1H NMR (CD3OD) δ: 1.15 (3H, s), 1.49-1.68 (6H, m), 2.25-2.40 (2H, m), 2.45-2.55 (2H, m), 3.45 (2H, s), 7.15 -7.30 (5H, m).
