Biotool Vita-Orange細胞活性檢測試劑是一種氧化還原反應的指示劑，在電子載體1-Methoxy PMS存在的條件下，利用活細胞中的脫氫酶催化四唑鹽WST-8生成甲臜染料（圖1），而且甲臜染料的生成量與活細胞的數(shù)量成線性關系（圖2）。該試劑的靈敏度高于MTT、XTT、MTS或WST-1等其他四唑鹽。
Biotool Vita-Orange細胞活性檢測試劑為一瓶即開即用的溶液，該試劑安全，無放射性，性價比高，為客戶提供方便、快速、可靠的使用體驗。

??細胞活力檢測: 細胞代謝活力與染料產生隨細胞活力的變化而變化。
??細胞因子實驗: 測定細胞因子誘導的增殖，恢復及擴增細胞。
??細胞毒性實驗: 有毒的化學物質導致的死亡的細胞對該檢測中顏色的變化沒有影響，只有活細胞能夠轉變顏色。Biotool Vita-Blue細胞活性檢測試劑本身無毒性，對細胞安全。

準備細胞

對于貼壁細胞，接種于培養(yǎng)瓶后使之貼壁并生長4–24小時，然后開始檢測。對于懸浮細胞，接種與培養(yǎng)瓶后可以立即檢測或24小時之內檢測。

實驗步驟

可選步驟：用測試化合物處理細胞24-72小時后進行該實驗。

1. 在細胞培養(yǎng)液中直接加入1/10體積的Biotool Vita-Blue細胞活性檢測試劑，充分混合，保證孔中藍色均一性，但避免氣泡產生。對96孔板，每100μl培養(yǎng)液加入10μl活性檢測試劑。
2. 于37°C培養(yǎng)箱中培養(yǎng)0.5-4小時至顏色變?yōu)槌壬?。應避免過夜培養(yǎng)。
3. 用熒光計數(shù)器測量在450nm處的吸收光。一般的OD值在0.5-2.5之間，典型的在0.8-1.5之間。
注意: 在添加Vita-Orange細胞活性檢測試劑的細胞培養(yǎng)基中，在450nm處會有輕微的自發(fā)吸收值。該熒光背景隨培養(yǎng)基、pH值、培養(yǎng)時間、暴露于光線的時間等因素的變化而變化。典型的吸收值在孵育2小時后為0.1-0.2。為了糾正背景，準備一個或多個無細胞的對照孔，計算其平均的熒光值，然后從試驗孔中扣除該值。
4.?可選步驟：在100μl細胞中加入10 μl 3% SDS 以終止反應，可在2天內讀取熒光值。

防范措施

1. 使用前請充分混勻，避免反復凍融。
2. 如果長時間孵育（過夜），請確保在試劑添加與孵育期間維持無菌狀態(tài)。細菌污染物能夠還原Biotool Vita-Orange細胞活性檢測試劑。
3. 在應用Biotool Vita-Orange細胞活性檢測試劑之后，還可以做其他細胞增殖實驗，如中性紅或結晶紫。
4. 確保包括合適的對照。

儲存條件

Biotool Vita-Orange細胞活性檢測試劑在避光條件下可在室溫保存6個月，2-8°C保存一年，-20°C及以下可保存更長時間。
