Biological Activity	Fully biologically active when compared to standard. The specific activity determined by an assay on anti-proliferation and anti-migration using endothelial cells in vitro and anti-angiogenesis in vivo is 5.5×10⁵IU/mg.
Physical Appearance	Sterile Filtered White lyophilized (freeze-dried) powder.
Formulation	Lyophilized from a 0.2μM filtered concentrated solution in 20mM NaAc, pH5.5, 4% mannitol.
Endotoxin	Less than 1EU/μg of rHuAngiostatin as determined by LAL method.
Reconstitution	We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute in sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0mg/ml. Stock solutions should be apportioned into working aliquots and stored at ≤-20℃. Further dilutions should be made in appropriate buffered solutions.

Background	Angiostatin, is a ~30kDa fragment of plasminogen that is encoded by the PLG gene in humans. It is produced, for example, bybautoproteolytic cleavage of plasminogen, involving extracellular disulfide bond reduction by phosphoglycerate kinase.bFurthermore, angiostatin can be cleaved from plasminogen by different metalloproteinases (MMPs), elastase, prostate-specific antigen (PSA), 13kDa serine protease, or 24kDa endopeptidase. Angiostatin is known to bind many proteins, especially to angiomotin and endothelial cell surface ATP synthase but also integrins, annexin II, C-met receptor, ng2 proteoglycan, tissuetype plasminogen activator, chondroitin sulfate proteoglycans, and CD26. It seems to involve inhibition of endothelial cell migration, proliferation and induction of apoptosis, but its mechanism of action is still unclear. Angiostatin is currently undergoing clinical trials for its use in anticancer therapy. Recombinant angiostatin is expressed in E. coli.

保存條件:
???????-20℃或更低溫度保存，至少一年有效。由于蛋白的每次凍融均會(huì)引起部分失活，所以首次配制成相應(yīng)濃度的儲(chǔ)存液后(請(qǐng)根據(jù)產(chǎn)品簡(jiǎn)介中Reconstitution一欄的信息配制儲(chǔ)存液)，須分裝后-20℃或更低溫度凍存，以避免反復(fù)凍融。
注意事項(xiàng):
???????由于有些塑料管壁對(duì)某些蛋白有較強(qiáng)的吸附作用，溶液中的蛋白很容易粘附在管壁上，并且粘附后的蛋白很難與管壁分離。而載體蛋白(Carrier protein，如0.1% BSA等)的主要作用是預(yù)先封閉塑料管壁上的蛋白結(jié)合位點(diǎn)，使細(xì)胞因子或重組蛋白不會(huì)粘附于管壁。所以一定要使用產(chǎn)品簡(jiǎn)介中Reconstitution一欄的信息配制儲(chǔ)存液。
