背景（Background）
FAP（也稱為seprase）是一種II型跨膜絲氨酸蛋白酶。質(zhì)膜和可溶性形式都表現(xiàn)出脯氨酸切割后的內(nèi)肽酶活性，對Ala/Ser-Gly-Pro-Ser/Asn/Ala共有序列有明顯的偏好。也降解明膠，熱變性I型膠原蛋白。還具有二肽基肽酶活性，優(yōu)選Ala-Pro、Ile-Pro、Gly-Pro、Arg-Pro和Pro-Pro。質(zhì)膜形式與DPP4、PLAUR或整合素結(jié)合，參與細(xì)胞外基質(zhì)（ECM）的細(xì)胞周蛋白水解，從而促進(jìn)細(xì)胞通過ECM的粘附、遷移和侵襲。通過降解短鏈蛋白多糖促進(jìn)膠質(zhì)瘤細(xì)胞通過腦實(shí)質(zhì)的侵襲。通過以絲氨酸蛋白酶活性無關(guān)的方式調(diào)節(jié)細(xì)胞增殖和存活，在黑素細(xì)胞中充當(dāng)腫瘤抑制因子。
