背景（Background）
HIV-1 p24 ELISA試劑盒基于ELISA夾心法，旨在測量CAR-T產(chǎn)品制備過程中樣本中的HIV-1 p23水平。它含有重組HIV-1 p24和一對抗重組因子的抗體，由ACROBiosystems提供。結(jié)果是通過與獲得的標(biāo)準(zhǔn)曲線平行的四參數(shù)邏輯斯諦曲線獲得的。驗(yàn)證結(jié)果表明，該試劑盒可用于定量測定HIV-1 p24濃度。

存儲（Storage）
1、未開封的試劑盒在收到后應(yīng)存放在2℃-8℃的溫度下。
2、在外包裝上找到有效期，不要使用超過有效期的試劑。
3、開封后請在30天內(nèi)使用。
原理（Assay Principles）
該檢測試劑盒采用標(biāo)準(zhǔn)的夾心ELISA，可快速檢測HIV-1 p24。該試劑盒由預(yù)包被的抗HIV-1 p24抗體微孔板、HIV-1p24標(biāo)準(zhǔn)品、生物素抗HIV-1 p24抗體、鏈霉抗生物素蛋白-HRP和緩沖液組成。
您的實(shí)驗(yàn)將包括6個簡單步驟：
a) Bring all reagents to room temperature(20℃-25℃) before use.
b) Add your sample to the plate and take the HIV-1 p24 as standard. The samples and standard are diluted by Dilution Buffer.
c) Wash the plate and add the Biotin-Anti-HIV-1 p24 Antibody diluted by Dilution Buffer to the plate.
d) Wash the plate and add the Streptavidin-HRP diluted by Dilution Buffer to the plate.
e) Wash the plate and add TMB.
f) Stop the substrate reaction by adding diluted acid. Absorbance (OD) is calculated by the absorbance at 450 nm minus the absorbance at 630 nm to remove background disturbance before statistical analysis. The OD Value reflects the amount of bound protein.
