名稱	Suramin Sodium Salt
描述	Suramin Sodium Salt (BAY-205) is a sodium salt form of suramin, a polysulphonated naphthylurea with potential antineoplastic activity. Suramin blocks the binding of various growth factors, including insulin-like growth factor I (IGF-I), epidermal growth factor (EGF), platelet-derived growth factor (PDGF), and tumor growth factor-beta (TGF-beta), to their receptors, thereby inhibiting endothelial cell proliferation and migration. This agent also inhibits vascular endothelial growth factor (VEGF)- and basic fibroblast growth factor (bFGF)-induced angiogenesis; retroviral reverse transcriptase; uncoupling of G-proteins from receptors; topoisomerases; cellular folate transport; and steroidogenesis.
激酶實(shí)驗(yàn)	The ATPase assay is performed in a 10 μL reaction mixture containing 20 mM Tris-HCl (pH 7.5), 1 mM DTT, 8 mM MgCl2, 5 μM M13 circular ssDNA, 2.5 μM RecA from the specified bacterial species and increasing concentrations of suramin. The reaction is initiated by the addition of 2 mM [α-32P]ATP, incubated for 30 min at 37°C and stopped by the addition of 25 mM EDTA[2].
體外活性	Suramin 在培養(yǎng)的HeLa細(xì)胞中抑制細(xì)胞增殖和DNA合成。40 μM suramin 完全抑制了SV40 DNA的復(fù)制。與DNA聚合酶δ（IC50=36 μM）相比，DNA聚合酶α對(duì)較低濃度的suramin更敏感（IC50=8 μM），而DNA聚合酶β對(duì)該化合物相對(duì)不敏感（IC50=90 μM）[1]。Suramin是細(xì)菌RecA蛋白DNA鏈交換和ATP酶活性的強(qiáng)效抑制劑。Suramin抑制RecA催化的LexA抑制因子的蛋白水解切割。Suramin的抑制作用機(jī)制涉及其瓦解RecA-單鏈DNA絲的能力[2]。Suramin是核酶DNA拓?fù)洚悩?gòu)酶II的強(qiáng)效抑制劑。Suramin抑制純化的酵母拓?fù)洚悩?gòu)酶II，其IC50約為5 μM[3]。
體內(nèi)活性	與對(duì)照組大鼠相比，經(jīng)Suramin處理后第21天的肺動(dòng)脈壓力、右心室肥大以及遠(yuǎn)端血管肌化指標(biāo)均有所降低。Suramin的治療能夠抑制PA-SMC的增殖，并減輕炎癥反應(yīng)與膠原蛋白的沉積[4]。
