LIF分子背景

白血病抑制因子（LIF）是一種白細(xì)胞介素6類細(xì)胞因子，是細(xì)胞中影響細(xì)胞生長(zhǎng)和發(fā)育的蛋白質(zhì)。白血病抑制因子具有多種功能，如膽堿能神經(jīng)元分化、控制干細(xì)胞多能性、骨和脂肪代謝、因子依賴性細(xì)胞系的有絲分裂和促進(jìn)體內(nèi)巨核細(xì)胞的產(chǎn)生。

LIF的去除推動(dòng)干細(xì)胞分化，但它們保留了增殖潛力或多能性。因此，LIF用于小鼠胚胎干細(xì)胞培養(yǎng)。有必要將干細(xì)胞保持在未分化狀態(tài)，但是胚胎干細(xì)胞的基因操縱允許LIF非依賴性生長(zhǎng)，特別是Nanog基因的過(guò)表達(dá)。人胚胎干細(xì)胞培養(yǎng)不需要LIF。
