艾塞那肽(exenatide)為胰高血糖素樣肽-1(GLP-1)類似物exendin-4的人工合成品，Exendin-4 (3-39)是Exenatide的片段多肽，由37個氨基酸組成，保留了Exenatide的大部分結(jié)構(gòu)。其結(jié)構(gòu)中的Asn殘基在高pH值和高溫下易發(fā)生脫酰胺反應(yīng)形成Asp，His和Trp殘基較易發(fā)生氧化，由Asp參與形成的肽鍵比其他肽鍵更易水解斷裂，除Gly外，其他所有氨基酸殘基易在堿催化下發(fā)生消旋反應(yīng)，在堿性條件下Thr殘基可通過β消除發(fā)生降解。因此在多肽合成過程中，需要實用合理的生產(chǎn)工藝。

Exendin-4 (3-39) is a fragment of Exenatide peptide, consisting of 37 amino acids , Retaining most of the structure of Exenatide. The Asn residues in its structure are prone to deamidation reaction to form Asp at high pH and high temperature. His and Trp residues are more prone to oxidation. The peptide bond formed by Asp is more prone to hydrolysis and cleavage than other peptide bonds, except for Gly , All other amino acid residues are prone to racemization reaction under base catalysis, and Thr residues can be degraded by β elimination under alkaline conditions. Therefore, in the process of peptide synthesis, a practical and reasonable production process is required.
