Amikacin sulfate is a polycationic, semisynthetic, bactericidal aminoglycoside. Amikacin was synthesized by acylation with the L-(-)-γ-amino-α-hydroxybutyryl side chain at the C-1 amino group of the deoxystreptamine moiety of kanamycin [K0047]. Amikacin is very active against most Gram-negative bacteria including gentamicin- [G0383] and tobramycin- [T2503] resistant strains. Amikacin enters the bacterial cell by binding to negatively charged components of the bacterial cell wall disrupting the overall architecture of the cell wall. The mechanism of action is disruption and inhibition of protein synthesis in the target bacteria by binding to the 30S ribosomal subunit.^1,2) In addition, amikacin is devoid of any chromophore and/or conjugated system prerequisite for UV and florescent light detection (FLD).
