體外研究活性 | BIBW2992不可逆地抑制EGFR/HER2,在體外作用于野生型EGFR ,L858R突變型EGFR, L858R/T790M 雙突變型EGFR,及HER2時IC50分別為0.5, 0.4, 10, 14 nM。在體外實驗中,BIBW2992不管作用于野生型EGFR/HER2還是突變型EGFR/HER2都顯示出強大的活力,與gefitinib作用于L858R突變型EGFR的活力差不多,但是要比gefitinib 作用于L858R-T790M雙突變型EGFR的活力大100倍。在體外,BIBW2992有效抑制所有實驗類型細胞的EGFR/HER磷酸化作用。比如人類扁平上皮癌A431細胞系表達的野生型EGFR, 鼠類NIH-3T3細胞轉染的野生型HER2 , 以及乳腺癌BT-474細胞系和胃癌NCI-N87細胞系表達的內源性HER2。[1] | |||

方法 | 細胞按1×104密度轉移到96孔板上,在無血清培養(yǎng)基上過夜培養(yǎng),用于EGFR磷酸化實驗。第二天添加包含BIBW2992的混合物,培養(yǎng)基在37oC溫育1小時。在室溫下用100 ng/ml EGF刺激10分鐘。細胞用冰凍PBS沖洗,然后每孔用120 μl HEPEX buffer萃取,然后在室溫下震蕩1小時。每孔中所有2×104細胞都用于HER2磷酸化實驗。在涂有鏈霉親和素的板上加用封閉液按1:100比例稀釋的抗EGFR的生物素和c-erb2/HER2腫瘤蛋白Ab-5-生物素。上述步驟得到的萃取物轉移到有抗體包被的孔中,室溫下溫育1小時。彩色顯影的測法見補充說明,吸光度為450納米。 |

體內研究活性 | 活體研究時按實驗對象體重,每千克口服20 mg BIBW2992持續(xù)25天,腫瘤產生明顯的衰退現(xiàn)象,通過腫瘤切片的免疫組織化學染色觀察發(fā)現(xiàn),EGFR和AKT的磷酸化作用收到抑制。因此,正如lapatinib和neratinib, BIBW2992是次代酪氨酸激酶抑制劑,不可逆地抑制人類EGFR2(HER2)和EGFR激酶。BIBW2992不僅能和原代酪氨酸激酶如erlotinib和gefitinib一樣有效抑制EGFR突變型,而且還能作用于抗erlotinib和gefitinib的EGFR。[1] | |
