體外研究活性 | 與DMSO處理的LGL T細胞相比,用5 μM Tipifarnib處理72小時,凋亡細胞百分率明顯高很多。使用健康捐贈者提供的T細胞,Tipifarnib按劑量依賴性降低IFNγ陽性細胞百分率。在沉淀物中,與DMSO相比,Tipifarnib降低激活型Ras的數(shù)量。[2]在體外,Tipifarnib濃度低于10 nM時,選擇性在克隆性骨髓增生異常綜合征(MDS)造血作用中產(chǎn)生毒性,當白細胞存在時這種作用更顯著。用Tipifarnib處理72小時,正常MDS前體表達出等量的DiOC3和膜聯(lián)蛋白V。[3] Tipifarnib和10 nM 4-OH-ICI 46474聯(lián)用,在有E2存在時,使IC50值降低8倍,從400 nM降低到50 nM。[4] |
