生物活性

Alloxan hydrate is a diabetogenic agent to induce diabetes. Alloxan hydrate is a proteasome inhibitor. Alloxan causes diabetes in experimental animals through its ability to destroy the insulin-secreting B-cells of the pancreas[1][2].

Alloxan hydrate inhibits the peptidase activities of the purified 26S and 20S proteasomes[1].

Alloxan hydrate 可用于動(dòng)物建模，構(gòu)建糖尿病模型。 Alloxan hydrate 給藥后導(dǎo)致小鼠血糖水平急劇升高，在 45 min 時(shí)達(dá)到峰值。給藥 2 小時(shí)后，血糖降低至接近低血糖水平，在 6 小時(shí)后達(dá)到最低血糖水平[3]。

Note
(1) Alloxan hydrate 溶解在鹽酸 (pH=5) 中，并在溶解后 10 分鐘內(nèi)進(jìn)行尾靜脈注射給藥。
(2) 通過(guò)眼眶穿刺進(jìn)行血液取樣，分別取 25 μL 和 250 μL 用于測(cè)定血糖和血漿胰島素水平。
(3) 血糖和血漿胰島素水平分別通過(guò)酶法和免疫法測(cè)定，血糖水平達(dá)到或超過(guò) 200 mg/100 mL 的小鼠被認(rèn)為患有糖尿病。
