L-Arginine ((S)-(+)-Arginine) is the substrate for the endothelial nitric oxide synthase (eNOS) to generate NO. L-Arginine is transported into vascular smooth muscle cells by the cationic amino acid transporter family of proteins where it is metabolized to nitric oxide (NO), polyamines, or L-proline. L-Arginine is a potent vasodilator, and can be used to induce experimental acute pancreatitis[1][2][3][4][5].

誘導(dǎo)實(shí)驗(yàn)性急性胰腺炎[4][5]

致病原理
L-Arginine 是內(nèi)皮型一氧化氮合酶 (eNOS) 產(chǎn)生 NO 的底物，能夠被代謝為成一氧化氮 (NO)、多胺或 L-脯氨酸，刺激炎癥反應(yīng)。L-Arginine 還可以選擇性破壞胰腺腺泡細(xì)胞，從而急性壞死性胰腺炎。
具體造模方法
Rat[4]: male • Wistar albino rats • 150 g
Administration: 250 mg, 500 mg, 750 mg every 100 g body weight • ip for single dose • starved for 15 hours
Mice[5]: male • ICT mouse • 4-week-old • 25-30 g
Administration: total 450 mg/100 g body weight • ip for 2 doses, with 1 hr interval • sacrificed at 72 hours
Note
1. 當(dāng)劑量超過(guò) 500 mg/100 g 體重時(shí)，將導(dǎo)致大部分大鼠在注射后數(shù)小時(shí)內(nèi)死亡；當(dāng)接受劑量低于 250 mg 時(shí)，誘導(dǎo)出現(xiàn)的胰腺病的癥狀會(huì)較輕[4]。
2. 文獻(xiàn)中溶解方法參考[5]：將 L-Arginine 粉末溶解于 0.9% 生理鹽水中，用 5 N 鹽酸調(diào)節(jié) pH 至 7，制得 L-Arginine 溶液。
造模成功指標(biāo)
外觀變化：體重減輕，活躍度較低，毛發(fā)較柔軟。
組織形態(tài)：胰腺水腫、損傷、萎縮，出現(xiàn)肉芽組織收縮和白堊色斑點(diǎn) (脂肪壞死位置)。
細(xì)胞變化：嗜堿性粒細(xì)胞減少和水泡化，細(xì)胞固縮、核破碎。胰泡細(xì)胞消失、凋亡，胰腺組織被單核細(xì)胞和成纖維細(xì)胞取代。
分子變化：血清淀粉酶水平增加，脂肪明顯壞死。
