    H2DCFDA (DCFH-DA) is a cell-permeable probe used to detect intracellular reactive oxygen species (ROS) (Ex/Em=488/525 nm)[1].

    (In Vitro)

    1.1 制備儲存液
    用 無水 DMSO 配制 5 mM 的 H2DCFDA 儲備液。
    注:H2DCFDA 儲存液建議分裝后于 -20 ℃ 或 -80 ℃ 避光保存。
    1.2 工作液的配制
    用預(yù)熱好的無血清細(xì)胞培養(yǎng)基或 PBS 稀釋儲存液,配制成 1-10 μM 的 H2DCFDA工作液。
    注:請根據(jù)實(shí)際情況調(diào)整 H2DCFDA 工作液濃度,且現(xiàn)用現(xiàn)配。
    2. 細(xì)胞染色(懸浮細(xì)胞)
    2.1 離心收集細(xì)胞,加入 PBS 洗滌兩次,每次 5 分鐘。細(xì)胞密度在 1×106 /mL。
    2.2 加入 1 mL 染料 工作液,室溫孵育 5-30 分鐘。
    2.3 400 g,離心 3-4 分鐘,棄去上清。
    2.4 加入 PBS 洗滌細(xì)胞兩次,每次 5 分鐘。
    2.5 用 1 mL 無血清培養(yǎng)基或 PBS 重懸細(xì)胞后,使用熒光顯微鏡或流式細(xì)胞儀進(jìn)行觀察。
    3. 細(xì)胞染色(貼壁細(xì)胞)
    3.1 將貼壁細(xì)胞培養(yǎng)于無菌蓋玻片上。
    3.2 從培養(yǎng)基中移出蓋玻片,吸除多余培養(yǎng)基。
    3.3 加入 100 μL 染料工作液,輕輕晃動使其完全覆蓋細(xì)胞,孵育 5-30 分鐘。
    3.4 吸去染料工作液,用培養(yǎng)基洗 2-3 次,每次 5 分鐘 ,使用熒光顯微鏡或流式細(xì)胞儀進(jìn)行 觀察。
    注:若需要用流式細(xì)胞儀檢測,需將細(xì)胞用胰蛋白酶消化重懸后再進(jìn)行染色。

      此方案可獲得 2.08 mg/mL的均勻懸濁液,懸濁液可用于口服和腹腔注射。

      1 mL 工作液為例,取 100 μL 20.8 mg/mL 的澄清 DMSO 儲備液加到 400 μL PEG300 中,混合均勻;再向上述體系中加入 50 μL Tween-80,混合均勻;然后再繼續(xù)加入 450 μL 生理鹽水 定容至 1 mL。

      生理鹽水的配制:將 0.9 g 氯化鈉,溶解于 ddH?O 并定容至 100 mL,可以得到澄清透明的生理鹽水溶液。

    ROS Measurements[1]
    For the detection of intracellular ROS level ,ROS-sensitive probe H2DCFDA is used. Adherent cells (ESCs, difESCs, eMSCs, HeLa, U118) are incubated with 5 μM staining solution in PBS in the dark for 30 min at 37°C, then harvested with 0.05% trypsin-EDTA solution, suspended in a fresh medium, and immediately analyzed with flow cytometer. Lymphocytes, both control and PHA-activated, are resuspended in PBS, incubated with 5 μM of H2DCFDA in the dark for 30 min at 37°C, and immediately analyzed. Along with the H2DCFDA probe, if indicated, ROS-insensitive modification of the fluorescent dye DCFDA is used. The staining procedure is the same as for the H2DCFDA[1].
