Rhodamine dyes are membrane-permeable cationic fluorescent probes that specifically recognize mitochondrial membrane potentials, thereby attaching to mitochondria and producing bright fluorescence, and at certain concentrations, rhodamine dyes have low toxicity to cells, so they are commonly used to detect mitochondria in animal cells, plant cells, and microorganisms[1].

體外研究
(In Vitro)

1. Rhodamine 123 工作液的配制
1.1儲備液的配制
取1 mg Rhodamine 123 溶于525 μL DMSO中，得到5 mM儲備液。
1.2 Rhodamine 123 工作液的配制
用無血清細(xì)胞培養(yǎng)基或PBS稀釋儲備液，得到1 -20 μM工作液。
注意：請根據(jù)實(shí)際情況調(diào)整 Rhodamine 123 工作液的濃度。
2.細(xì)胞染色
2.1 懸浮細(xì)胞 (6孔板)
a.1000g 4℃離心3-5分鐘，棄上清。PBS洗2次，每次5分鐘。細(xì)胞密度為1×106/mL。
b.加入1 mL工作液，室溫孵育5-30分鐘。
c.400g 4℃離心3-4分鐘，棄上清。
d.PBS洗滌2次，每次5分鐘。
e.重懸細(xì)胞用無血清細(xì)胞培養(yǎng)基或 PBS。熒光顯微鏡或流式細(xì)胞儀觀察。
2.2 貼壁細(xì)胞
a.在無菌蓋玻片上培養(yǎng)貼壁細(xì)胞。
b.從培養(yǎng)基中取出蓋玻片并吸出多余的培養(yǎng)基。
c .加入100 μL工作液，輕搖使之完全覆蓋細(xì)胞，然后室溫孵育30-60分鐘。
d.用培養(yǎng)基洗滌2次，每次5分鐘。熒光顯微鏡或流式細(xì)胞儀觀察。
注：若采用流式細(xì)胞儀檢測，細(xì)胞需重懸后再染色。

Measurements are made at room temperature with continuous stirring of the mitochondrial suspension using spectrophotometer equipped with a magnetic stirrer with fluorescent cation R123 as probe. Excitation and emission wavelengths are 503 nm and 527 nm, respectively. The incubation medium is the respiration buffer. R123 and sodium pyruvate are added to final concentrations of 50 nM and 10 mM, respectively. Isolated mitochondria maintain a steady membrane potential (±5%) throughout the duration of the recording[1].
