Penitrem A is an indole diterpene neurotoxic alkaloid produced by Penicillium, acts as a selective BK channel antagonist with antiproliferative and anti-invasive activities against multiple malignancies. Penitrem A increases the spontaneous release of endogenous glutamate, gamma-aminobutyric acid (GABA) and aspartate from cerebrocortical synaptosomes, and induces tremorgenic syndromes in animals[1][2].

致病原理
Penltrem A 中毒的生物化學(xué)效應(yīng)包括脂質(zhì)增加，但肝臟中的 DNA 和糖原減少，以及血清酶和厭氧糖酵解產(chǎn)物的升高。這些血清變化被認(rèn)為與毒素引起的過度肌肉活動(dòng)有關(guān)[1]。
Penltrem A 引起的震顫可能用與突觸前作用和抑制中間神經(jīng)元來解釋[2]。
具體造模方法：
小鼠：ICR 成年 • 雄性 • 體重 38-46 g[1]
給藥：10 mg/kg • 皮下注射 • 單次劑量，持續(xù) 72 小時(shí)或每 3 天一次，持續(xù) 18 天[1]
大鼠：裸鼠 Wistar • 雄性 • 體重 170 和 290 g[2]
給藥：腹腔注射 0.1 mL/100 g 或 腦室內(nèi)注射 3 μg/μL • 腹腔注射或腦室內(nèi)注射 • 單次劑量，持續(xù) 1, 3, 7 天[2]

(1) 在實(shí)驗(yàn)開始之前，動(dòng)物飼養(yǎng)設(shè)施的溫度維持在 21℃，濕度 50%，采用 12 小時(shí)的明暗周期。給老鼠提供 Agway RMH 3000 顆粒飼料和隨意飲水[1][2]
(2) Penltrem A 溶解在玉米油中，使得適當(dāng)?shù)膭┝繛?2.5 mL/kg，并通過皮下注射給藥[1]。
(3) 對(duì)于腹腔注射，Penltrem A 溶解在聚乙烯醇 300 (PEG 300) 中，劑量為 0.1 mL/100 g 體重。對(duì)于腦室內(nèi)給藥，Penltrem A 溶解在 75% 的 PEG 300 和 25% 的生理鹽水中，濃度為 3 μg/μL。[2]
