    Rhodamine dyes are membrane-permeable cationic fluorescent probes that specifically recognize mitochondrial membrane potentials, thereby attaching to mitochondria and producing bright fluorescence, and at certain concentrations, rhodamine dyes have low toxicity to cells, so they are commonly used to detect mitochondria in animal cells, plant cells, and microorganisms[1].

    1.TMRM 工作液的配制
    1.1 儲(chǔ)備液的配制
    取 1mg TMRM 溶于 339 uL DMSO 中，得到 5 mM 儲(chǔ)備液。
    1.2 TMRM 工作液的配制
    用無血清細(xì)胞培養(yǎng)基或 PBS 稀釋儲(chǔ)備液，得到 1 -20 μM 工作液。
    注意：請(qǐng)根據(jù)實(shí)際情況調(diào)整 TMRM 工作液的濃度。
    2. 細(xì)胞染色
    2.1 懸浮細(xì)胞 (6 孔板)
    a.1000g 4℃ 離心 3-5 分鐘，棄上清。PBS 洗 2 次，每次 5 分鐘。細(xì)胞密度為 1×106/mL。
    b. 加入 1 mL 工作液，室溫孵育 5-30 分鐘。
    c. 400g 4℃ 離心 3-4 分鐘，棄上清。
    d. PBS 洗滌 2 次，每次 5 分鐘。
    e. 重懸細(xì)胞用無血清細(xì)胞培養(yǎng)基或 PBS。熒光顯微鏡或流式細(xì)胞儀觀察。
    2.2 貼壁細(xì)胞
    a. 在無菌蓋玻片上培養(yǎng)貼壁細(xì)胞。
    b. 從培養(yǎng)基中取出蓋玻片并吸出多余的培養(yǎng)基。
    c . 加入 100 μL 工作液，輕搖使之完全覆蓋細(xì)胞，然后室溫孵育 30-60 分鐘。
    d. 用培養(yǎng)基洗滌 2 次，每次 5 分鐘。熒光顯微鏡或流式細(xì)胞儀觀察。

    Cultures are exposed to Millipore-filtered solutions (0.22 μm) containing TMRM Perchlorate for 1 hr at 37°C (except the experiment involving different durations of exposure to TMRM Perchlorate). After treatment, solutions are removed and growth media reapplied under sterile conditions, and cultures are post-incubated for 18 hours at 37°C (except for the experiment involving analysis at different time points after exposure). Cells are then stained with 2 mg/mL bisbenzimide for 20 min at room temperature. Coverslips are subsequently washed in saline and imaged using 2P microscopy. Apoptotic cells are identified as brightly fluorescent nuclei under UV excitation indicating DNA fragmentation. Cell survivability is calculated as the percentage of live, unstained cells (±SD) in five microscopic fields per treatment[1].
