      致病原理
      MPTP 可以自由通過血腦屏障進入大腦，在大腦內(nèi)，MPTP 被星形膠質(zhì)細胞中的單胺氧化酶B (MAO-B) 代謝轉(zhuǎn)化為 MPP+，這是其活性、有毒性的形式。MPP+ 被多巴胺神經(jīng)元通過多巴胺轉(zhuǎn)運體 (DAT) 攝取，阻斷線粒體電子傳遞鏈中的復合體 I (Complex I)，引發(fā)氧化應(yīng)激和線粒體破裂，最后導致神經(jīng)元凋亡。在帕金森病中，主要是多巴胺產(chǎn)生的部位“黑質(zhì)-紋狀體系統(tǒng)”的黑質(zhì)部分神經(jīng)元喪失導致病情。由于 (MPTP) 的毒性作用，會導致黑質(zhì)中的多巴胺神經(jīng)元死亡，從而誘發(fā)出類似帕金森病的癥狀。
      具體造模方法：
      小鼠：C57BL/6 • 雄性 • 8-12 周齡 (處理：2 周), 高齡小鼠會更易敏感。
      給藥方式：
      Acute model：14-20 mg/kg • 腹腔注射 • 每天 4 次，間隔兩小時
      Sub-acute model：20-30 mg/kg • 腹腔注射 • 每天一次，連續(xù) 5 天
      Note
      1. MPTP Hcl 溶解在生理鹽水中，現(xiàn)用現(xiàn)配。
      2. 注射后，肉眼可以注意觀察小鼠是否有活動性減弱、走路踉蹌，抽搐、炸毛，排尿變多等表現(xiàn)，這種行為可能可以持續(xù) 24-48 小時，此后小鼠表現(xiàn)基本正常。
      3. MPTP 通常以 MPTP 鹽酸鹽的形式售賣。 MPTP 鹽酸鹽分子量為209.7。 因此，在制備注射液時建議考慮到鹽酸鹽 (HCl)的存在。HCl 的分子量為 35.4，占 MPTP 的 17%。 因此，如果要制備 20 mg/kg 劑量的 MPTP，則 MPTP 鹽酸鹽給藥劑量為 20 mg kg* 1.17% = 23.4 mg/kg。
      4. 如果在 1 天內(nèi)進行多次注射，最好交替進行兩側(cè)注射。如果每天注射，應(yīng)保證在同一時間點。每次注射前均需給小鼠稱重，調(diào)整給藥體積。
      5. 造模小鼠不一定會出現(xiàn)帕金森病行為缺陷，小鼠個體差異性也較大,造模成功率一般難達 100%。因此 MPTP 小鼠研究中要監(jiān)測與神經(jīng)膠質(zhì)增生相關(guān)的黑質(zhì)紋狀體損傷。
      6. 藥物劑量高/小鼠體重小于 22 g/不同批次藥物混用/小鼠沒有提前適應(yīng)/動物房太冷 均有可能會導致小鼠死亡，每組動物數(shù)目建議增大，必要時根據(jù)預(yù)實驗情況調(diào)整劑量。
      造模成功指標
      1) 黑質(zhì)紋狀體損傷: 造模成功后黑質(zhì)和紋狀體區(qū)域酪氨酸羥化酶 (Tyrosine hydroxylase) 減少 (IHC, IF, WB 等方法都可以);
      2) 其他指標：大腦神經(jīng)遞質(zhì) (DA, DOPAC, 5-HT, HVA, etc.) 減少 (可通過HPLC 檢測);
      3) 黑質(zhì)紋狀體小膠質(zhì)細胞 (IBA1+ cells) 和星形膠質(zhì)細胞 (GFAP+ cells) 激活, 黑質(zhì)區(qū) α-syn 聚集體數(shù)量增加。

      For the preparation of the LPS rat model and the MPTP mouse model, the treatments of the animals are performed. Briefly, adult rats receive unilateral injections of LPS (0.5?μL of 10?μg/μL diluted in 0.9% saline) into the medial forebrain bundle (MFB) at the following coordinates, AP-4.2?mm, L 1.5?mm, and V 7.8?mm, and into the contralateral side with the same volume of 0.9% saline. Adult mice are administered intraperitoneal injections of MPTP of 25?mg/kg per day for five continuous days, and the same volume of saline is injected as a control. All the animals are sacrificed at week 1, 2, 3, or 4 after the LPS or MPTP injections. The brain samples are collected for the subsequent immunohistochemistry and western blot experiments.
