對(duì)照組與實(shí)驗(yàn)組的準(zhǔn)備：實(shí)驗(yàn)需包含平行的陰性、陽(yáng)性和實(shí)驗(yàn)組。實(shí)驗(yàn)組：加入含有目標(biāo) GSH 效應(yīng)物 (如 200 μM H2O2) 的 200 μL 細(xì)胞培養(yǎng)基。
在 37°C、5% CO2 下孵育培養(yǎng)板過(guò)夜，具體時(shí)間可根據(jù)常規(guī)實(shí)驗(yàn)方案調(diào)整。加入 Monochlorobimane (mBCl) ：終濃度為 20-100 μM，使用 1 mM 的工作溶液。mBCl 儲(chǔ)存于 -20°C，實(shí)驗(yàn)前用 0.1 M PBS (pH 7.0) 稀釋制備。實(shí)驗(yàn)中最終 DMSO 濃度低于 0.2% (v/v) 。
在暗處 25°C 孵育 ~2 小時(shí)。通過(guò)離心去除染料和處理液 (700 × g, 5 分鐘) ，加入 200 μL 檢測(cè)緩沖液，繼續(xù)檢測(cè)步驟。檢測(cè)細(xì)胞內(nèi) GSH：熒光板檢測(cè)：貼壁細(xì)胞可直接在培養(yǎng)板中讀取熒光；懸浮細(xì)胞需將 200 μL 細(xì)胞懸液加入不透明的白板，并記錄熒光 EX/EM=380/465±20 nm (藍(lán)色) 。

Monochlorobimane (mCB) is added to the second half liver tissue homogenate to a final concentration of 100 mM along with glutathione S-transferase (1 U/mL) obtained from equine liver; the homogenate is then allowed to incubate at room temperature for 30 min. The Glutathione (GSH)-Monochlorobimane adduct is measured in a microtiter reader[1].
