Nicotinamide是一種水溶性維生素，是NAD 和 NADP的活性成分，也是一種sirtuins抑制劑。

體外研究(In Vitro)
體外研究活性	Nicotinamide強(qiáng)烈抑制酵母（SIR2突變）沉默，增加的rDNA重組，并縮短復(fù)制壽命。Nicotinamide廢除沉默，導(dǎo)停滯致在G1期的細(xì)胞中Sir2離域，這表明沉默的異染色質(zhì)需要不斷的Sir2的活性。^[1] Nicotinamide導(dǎo)致胎兒細(xì)胞中DNA含量增加2倍，胰島素含量增加3倍。Nicotinamide誘導(dǎo)人胎兒胰島細(xì)胞的分化和成熟。^[2] Nicotinamide通過轉(zhuǎn)換去乙?；突鶊F(tuán)交換來調(diào)節(jié)乙?；?。Nicotinamide切換定量為Sir2s從Archeaglobus fulgidus（Sir2Af2），釀酒酵母（Sir2p）和小鼠（Sir2alpha）。^[3] Nicotinamide選擇性降低的特定磷酸化tau蛋白（Thr231），和阿爾茨海默氏病的轉(zhuǎn)基因小鼠的微管解聚有關(guān)，類似于對SIRT1的抑制。在阿爾茨海默氏病的轉(zhuǎn)基因小鼠中，Nicotinamide也極大地增加了乙酰化的α-微管蛋白（SIRT2的一級底物）和MAP2c，這兩者都與增加的微管的穩(wěn)定性有關(guān)。^[4] Nicotinamide促進(jìn)DNA的完整性，并保持磷脂膜不對稱性，以防止細(xì)胞炎癥，細(xì)胞吞噬功能和血管栓塞。Nicotinamide既防止也逆轉(zhuǎn)神經(jīng)和血管細(xì)胞的損傷。^[5]

	方法	將HaCaT細(xì)胞于含10%FBS的DMEM培養(yǎng)基中，置于在37℃、5% CO2培養(yǎng)箱中培養(yǎng)。對于NAD(P)調(diào)節(jié)實驗，將細(xì)胞培養(yǎng)于含10%透析FBS的DMEM培養(yǎng)基中，加入33 μM nicotinamide或不加入。7天或14天后，統(tǒng)計細(xì)胞數(shù)目，用0.1 μM 6-diazo-5-oxo-L-norleucine (DON)檢測細(xì)胞對谷酰胺酶抑制的敏感性。

體內(nèi)研究(In Vivo)
體內(nèi)研究活性	在小鼠中，通過腹腔注射，注射100-500 mg/kg的nicotinamide到體內(nèi)將產(chǎn)生雙相消除，其半衰期隨著劑量的變化而變化。初始半衰期將顯著增加，從0.8遞增至2h；而終末半衰期從3.4增至5.6 h。而其在體內(nèi)的消除率則只在最高濃度下發(fā)生顯著減少，從0.3 L/kg/h減少至0.24 L/kg/h。體內(nèi)峰值濃度也隨著給藥劑量發(fā)生增加，變化范圍為1000-4800 nmol/ml。通過腹腔注射的藥物生物利用度與靜脈注射相比，接近100%^[6]。
