體外研究活性 | Apigenin通過與三磷酸腺苷(ATP)競爭而抑制PKC。Apigenin也降低TPA誘導(dǎo)的細胞蛋白的磷酸化水平,并抑制TPA誘導(dǎo)的c-jun和c-fos表達。Apigenin對轉(zhuǎn)化的v-H-ras表型的NIH3T3細胞具有反轉(zhuǎn)作用。[1]在中國倉鼠卵巢細胞中,Apigenin能夠抑制硝基芘誘導(dǎo)的毒性基因引起的突變。在小鼠巨噬細胞中,Apigenin抑制LPS誘導(dǎo)的環(huán)氧合酶-2 和一氧化氮合成酶-2的活性和表達。Apigenin被報道具有抑制蛋白激酶C,分裂素激活的蛋白激酶(MAPK),C3HI小鼠胚胎成纖維細胞的轉(zhuǎn)化以及v-Ha-ras-轉(zhuǎn)化的NIH3T3細胞中下游致癌基因的活性。 Apigenin阻斷過氧物酶體增殖調(diào)控的激酶(ERK),離體肝細胞中的MAPK。Apigenin可以下調(diào)Na+/Ca2+-交換器的表達,該蛋白是大鼠心肌細胞鈣離子外排的一個重要蛋白。在表皮細胞和成纖維細胞中,Apigenin通過抑制p34 (cdc2)激酶活性引起G2/M and G0/G1可逆阻滯,并伴隨p53蛋白質(zhì)穩(wěn)定性增加。在培養(yǎng)的人類內(nèi)皮細胞中,Apigenin能夠有效抑制TNFα誘導(dǎo)的細胞粘附分子-1的上調(diào)。在人類卵巢癌細胞中,Apigenin通過PI3K/Akt/p70S6K1和HDM2/p53信號通路抑制HIF-1α 和 VEGF表達[2]在人類角質(zhì)細胞中,Apigenin通過抑制MAPK信號轉(zhuǎn)導(dǎo)并降低API轉(zhuǎn)錄因子水平抑制分化。Apigenin也會抑制人類角質(zhì)細胞的增殖。[3] | |||

體內(nèi)研究活性 | 在卵清蛋白免疫的BALB/C小鼠中,Apigenin下調(diào)IL-4的產(chǎn)生。Apigenin通過抑制腫瘤細胞和內(nèi)皮細胞的相互作用抑制黑色素瘤的肺轉(zhuǎn)移。在雌性小鼠中,Apigenin引起子宮重量和整個子宮中雌激素受體(ER)-α濃度顯著增加,通過雌激素受體β1抑制前列腺癌和乳腺癌細胞生長。Apigenin抑制前列腺移植瘤中IGF-I水平,增加在血液循環(huán)中結(jié)合IGF-I 蛋白的IGFBP-3含量。[2] Apigenin (12.5 毫克/千克)增加成年小鼠海馬區(qū)齒狀回區(qū)域細胞的增殖。[4] |
