體外研究活性

Z-VAD-FMK (10 μM)作用于THP.1細(xì)胞，抑制細(xì)胞凋亡。Z-VAD-FMK(10 μM)作用于THP.1細(xì)胞裂解液，抑制PARP蛋白酶活性的激活。Z-VAD-FMK (10 μM)作用于完整THP.1和 Jurkat細(xì)胞，抑制CPP32加工。[1]Z-VAD-FMK(50 μM)與Camptothecin共處理HL60細(xì)胞，廢除Camptothecin處理的細(xì)胞凋亡形態(tài)。Z-VAD-FMK(50 μM)作用于HL60細(xì)胞，抑制Camptothecin誘導(dǎo)的DNA分裂。[2]Z-VAD-FMK (50 μM)作用于S2細(xì)胞，提高轉(zhuǎn)染細(xì)胞的生存百分?jǐn)?shù)從26％提高到63％。[3]Z-VAD-FMK(>100 μM)作用于人類嗜中性粒細(xì)胞，增強(qiáng)TNFα-誘導(dǎo)的中性粒細(xì)胞凋亡，較低濃度(1-30 μM)時(shí)完全抑制TNFα刺激的細(xì)胞凋亡。[4]Z-VAD-FMK (10 mM) 作用于早期角膜基質(zhì)細(xì)胞，抑制細(xì)胞凋亡。[5]

方法

為了確定Z-VAD-FMK的效力，在常氧條件下，對(duì)三種人源顆粒細(xì)胞系(GC1a, HGL5, COV434)進(jìn)行處理，處理48小時(shí)。為了模擬卵巢片段抑制后的缺氧狀態(tài)，用低氧（1% O2）狀態(tài)下，用無血清培養(yǎng)基培養(yǎng)細(xì)胞，處理以Z-VAD-FMK。用WST-1試驗(yàn)檢測細(xì)胞的代謝活性。通過FACS檢測細(xì)胞活力，RT-qPCR和WB檢測凋亡相關(guān)分子的表達(dá)。

有研究表明，體內(nèi)Z-VAD-FMK的給藥不具有毒性，在動(dòng)物模型中可抑制凋亡。在小鼠中，腹腔注射HK-GBS可引起早產(chǎn)，而預(yù)處理以Z-VAD-FMK可延遲早產(chǎn)現(xiàn)象。在OVA-sensitized的小鼠中，z-VAD-fmk的處理可抑制過敏原誘導(dǎo)的白細(xì)胞浸潤。在OVA致敏前全身性注射z-VAD-fmk可減少OVA-sensitized/challenged小鼠中炎性細(xì)胞的積累、黏液的高分泌以及Th2細(xì)胞因子的釋放。處理以z-VAD-fmk可阻止晶狀體上皮細(xì)胞、角質(zhì)細(xì)胞的終末分化；單核細(xì)胞向巨噬細(xì)胞的分化；以及紅系造血祖細(xì)胞的分化。z-VAD-fmk可減緩過敏原誘導(dǎo)的呼吸道炎癥以及高敏性、阻止后續(xù)T細(xì)胞的激活[7]。
